It is best to do this all on a new layer and then freezing all other layers after you make all of your fillable regions. Once this is done you can export it as a dxf and when you open it in the vector program, I use CorelDraw, you can select the shapes and apply fills.

I’m if there were an way to get it as SVG, that would make it more manageable but maybe that’s a silly idea as well : ( printing to PDF will give you a traansparent background. Then open up the PDF with a photo editing software and save it as a PNG.

You will probably need to find a different printer driver to use from within AutoCAD; as far as I’ve ever been able to determine, you can’t make the background transparent when printing to PNG or any other format from AutoCAD. If someone can show me I’m wrong, I’d love to see it!
