I have a weird problem with SharePoint 2010 site permission which just started happening the other day and I can’t find a solution. If I manually grant/add a user access to a site through site actions > site permissions. The user can’t access the site even though it says they have access.
Everyone except external users That group captures all internal users only. Internal means they have been assigned your domain name when set up in Office 365. It does not matter whether the user has been granted an Office 365 license. In any instance, the user is part of the group.
How can I remove a user from my SharePoint account?
Sign in to https://admin.microsoft.com as a global or SharePoint admin. In the left pane, select Users > Guest users. Select Delete a user. Select the user, then Select, and then select Delete. Once the above action has been completed, ensure the account is completely removed from the site collection you are sharing with the user.

No, the default permission levels are not working because when I put someone in the built-in Visitor’s group, which has read-only rights, they cannot access the SharePoint site. I am trying to understand why they can’t access the site unless I put them into the built-in Member’s group.
When do you need permission to access a site?

When I put a user in a Read-Only group I created, which has Read permissions, the user gets “access denied” to the site. If I make the user a member of the O365 group and they are in one of these 2 SharePoint groups with Read-only rights, they still get access denied to the site.
